The NAD system is increasingly becoming the focus of scientific research concerning cellular energy, healthy aging (longevity), and the function of mitochondria. This doesn't just involve a single molecule but a complex interplay of various precursors & forms of NAD, which assume different tasks within the mitochondrial energy metabolism in the human body.



Mitochondria are considered the powerhouses of our cells and produce most of the cellular energy. From a biochemical perspective, this process does not occur in a single step but through several successive metabolic pathways. The NAD system accompanies this energy production along various biological stations.



A starting point in natural metabolism is Vitamin B3 (Niacin), from which the body can form NAD⁺, among other things, via Nicotinamide Riboside (NR). NAD⁺ is the oxidized form of the NAD system and acts as an electron acceptor in energy metabolism. In the citric acid cycle – a central part of mitochondrial energy production – energy-rich electrons are transferred to NAD⁺. Calcium-alpha-ketoglutarate (CAK) is a component of this metabolic pathway and is involved in reactions of the citric acid cycle. The absorption of electrons leads to the formation of NADH – the reduced and electron-loaded form. NADH then transports these electrons to the electron transport chain, where coenzyme Q10 takes over further electron transport. The energy released in this process is finally used for the formation of ATP – the universal energy currency of our cells.



Where energy is produced, reactive oxygen species (ROS) also arise as a natural accompanying process. In larger quantities, they can contribute to oxidative stress. Therefore, the body has its own antioxidant defense system that continuously neutralizes these reactive oxygen species. Only the interaction of energy production & the body's own defense system enables a balanced mitochondrial metabolism.

What is the NAD system, simply explained?

The term "NAD System"describes the interplay of NAD⁺, NADH, as well as the precursors and metabolic pathways involved in the formation and regeneration of NAD. The focus is on the continuous cycle between NAD⁺ and NADH: In the citric acid cycle, NAD⁺ accepts electrons and is converted to NADH. NADH then transports these electrons to the mitochondrial respiratory chain, where they are released. In this process, NADH is converted back to NAD⁺ – and the cycle begins again.

Biochemically, this redox system forms a central component of mitochondrial energy metabolism and is also involved in numerous other metabolic processes. Due to its importance and age-related changes in NAD metabolism, it is currently the focus of healthy aging and longevity research.
